Metode sampling yang digunakan yaitu metode acak. Penelitian ini dilaksanakan pada lima stasiun yang berbeda selama tiga kali waktu sampling dimana stasiun 1 merupakan hulu sungai, stasiun 2 merupakan representasi pengaruh hulu dan pasang surut, stasiun 3 merupakan muara sungai, stasiun 4 di dekat lokasi pertambakan, dan stasiun 5 di dekat aktivitas nelayan. Kelimpahan fitoplankton di sungai Jali dan sungai Lereng berkisar 3.503 – 12.165 ind/l berdasarkan kelimpahan fitoplankton menunjukkan bahwa perairan sungai Jali dan sungai Lereng tergolong tingkat kesuburannya sedang atau mesotrofik. Jenis fitoplankton yang ditemukan pada sungai Jali dan sungai Lereng Purworejo selama pengamatan terdiri dari 3 kelas yaitu kelas Bacillariophyceae , Chlorophyceae dan Cyanophycae serta terdiri dari 20 genera. Jenis zooplankton terdiri dari 2 filum yaitu filum Protozoa dan filum Crustacea, serta terdiri dari 8 genera. Kandungan nitrat berkisar 0,3 – 1,6 mg/l dan kandungan fosfat berkisar 0,04 – 0,7 mg/l. Nitrat memiliki korelasi sebesar 0,619 terhadap kelimpahan fitoplankton, hal tersebut berarti nitrat memiliki hubungan positif kuat dan fosfat memiliki korelasi sebesar 0,596 terhadap kelimpahan fitoplankton, hal tersebut berarti fosfat memiliki hubungan positif sedang. Water quality alteration has tight relationship with water potency, refers to phytoplankton abundance and composition. Phytoplankton is one of biology paramater that can be an indicator to evaluate water quality and fertility. This study aims to recognize water fertility based on the abundance and composition of phytoplankton and zooplankton in Jali River and Lereng River Purworejo and the relation between nitrate phosphate and phytoplankton abundance in Jali River and Lereng River Purworejo. Random sampling method are used in this observation. There are 5 different stations in 3 sampling periods. Station 1 is the upper course of river, station 2 is the representation of upper course and tide, station 3 is river mouth, station 4 is near to local fish pond and station 5 is near to fishing activity. Abundance of phytoplankton in Jali River and Lereng River value 3.503 – 12.165 ind/l based on phytoplankton abundancy it is showing that water quality of Jali River and Lereng River categorized in moderated fertility levels or mesotrofik. There are 3 classes of phytoplankton that found in study site. They are Bacillariophyceae, Chlorophyceae and Cyanophyceae. Then, consisting of 20 genera. 2 kinds of zooplankton are Protozoa filum and Crustacea filum, consisting 8 genera. Nitrate value is 0,3 – 1,6 mg/l and phosphate value is 0,04 – 0,7 mg/l.
