Metode yang digunakan adalah metode studi kasus yang bersifat deskriptif, sedangkan metode pengambilan sample yang digunakan dalam penelitian ini adalah dengan menggunakan metode sistematik sampling. Stasiun sampling terdiri dari 3 stasiun. Metode kerja meliputi pengambilan sampel fitoplankton dan sampel air untuk diukur kandungan unsur hara serta dilakukan pula pengukuran parameter pendukung seperti arus, pH, DO, kedalaman, dan kecerahan. Sampel fitoplankton diidentifikasi dan dihitung keanekaragaman, keseragaman, dominasi, dan kelimpahan. Hasil penelitian menunjukkan kandungan Nitrat berkisar 0,84-1,21 mg/l dan Fosfat berkisar 0,31-0,45 mg/l. Fitoplankton yang telah ditemukan 19 genera dari 3 kelas yaitu Bacillariphyceae 5 genera, Chlorophyceae 11 genera dan Cyanophyceae 3 genera, dengan genera yang banyak ditemukan yaitu Nitzchia sp. Kelimpahan fitoplankton yang diperoleh berkisar anatara 407,632-704,864 ind/l, kelimpahan fitoplankton tertinggi pada stasiun 2 yaitu 704,864 ind/l dan terendah pada stasiun 1 yaitu 407,632 ind/l. Nilai H’ yaitu 1,496 – 1,579 maka pencemaran di sungai Gambir tergolong Sedang. Berdasarkan uji statistik, ada korelasi positif antara unsur hara ( nitrat dengan kelimpahan fitoplankton, nilai r : 0,939 dan fosfat dengan kelimpahan fitoplankton, nilai r : 0,647). Gambir rivers are in fields and around densely populated residential area Banyumanik, it is estimated that many domestic waste that goes into the water bodies that will affect the fertility waters and p ollution that can be seen from the indicators of nutrients and phytoplankton abundance. The purpose of this study was to determine the content of nutrients (nitrates and phosphates), knowing the abundance of phytoplankton and the level of pollution in the River Gambir. This research was conducted in December 2014 - January 2015 in Gambir River, Tembalang Semarang. The method used is the method of descriptive case studies, while the sampling method used in this research is to use a systematic method of sampling. Sampling station consists of three stations. Working methods include sampling of phytoplankton and water samples to measure the nutrient content and also conducted support parameter measurements such as flow, pH, DO, depth, and brightness. Samples of phytoplankton were identified and counted diversity, uniformity, domination, and abundance. The results showed nitrate content ranged from 0.84 to 1.21 mg/l and Phosphate ranging from 0.31 to 0.45 mg/l. Phytoplankton has found 19 genera of 3 classes, namely Bacillariphyceae 5 genera, 11 genera Chlorophyceae and Cyanophyceae 3 genera, the genera are found that Nitzchia sp. The abundance of phytoplankton were obtained ranging from 407.632 to 704.864 anatara ind/l, the highest phytoplankton abundance at station 2 is 704.864 ind/l and the lowest at station 1 is 407.632 ind/l. The H’value is 1.496 to 1.579, the level of pollution in the river Gambir is moderate polluted classified.
