Metode yang digunakan dalam penelitian ini adalah metode studi kasus. Makrozoobenthos yang diperoleh selama penelitian di muara sungai Wedung Kabupaten Demak terdiri dari 3 kelas yaitu Polychaeta, Gastropoda, dan Bivalvia. Kelimpahan makrozoobenthos terbesar terdapat pada stasiun II sebesar 4889 ind/m 3 , sedangkan kelimpahan terendah terdapat pada stasiun III sebesar 341 ind/m 3 . Kandungan bahan organik stasiun I berkisar antara 11,81 – 15,45% (sedang), stasiun II berkisar antara 12,72 – 17,57% (tinggi), dan stasiun III berkisar antara 9,5 – 16,44% (sedang). Hasil uji regresi antara kelimpahan makrozoobenthos dengan kandungan bahan organik diperoleh nilai koefisien determinasi (R 2 ) sebesar 0,46. Koefisien korelasi (r) sebesar 0,678 yang menunjukkan bahwa hubungan antara kedua variable tersebut cukup. One of the caused was that natural pollution such as the organic matter that will have an impact on the lives of macrozoobenthos. The purpose of this study was to determine the composition and abundance of macrozoobenthos and to know the relationship between organic matter content of sediment with macrozoobenthos abundance in estuarine Wedung. The research was conducted in April to June 2013. This study was conducted at 3 stations. The research material was macrozoobenthos and Wedung estuarine substrate. The method used in this study was case study methods. Macrozoobenthos obtained during research in Wedung estuaries, Demak consists of three classes which were Polychaeta, Gastropoda, and Bivalve. The greatest abundance found at station II was 4889 ind/m 3 , while the lowest abundance at station III was 341 ind/m 3 . The content of organic matter in station I ranges from 11,81 to 15,45% (moderate), station II ranges from 12,72 to 17,57% (high), and station III ranges from 9,5 to 16,44% (moderate ). Results of regression between macrozoobenthos abundance with organic content obtained coefficient of determination (R 2 ) of 0,46.
