Penggunaan sedimen sebagai acuan dianggap lebih tepat, oleh karena sedimen relatif diam tidak bergerak seperti air sungai, sehingga lebih refresentatif dan mewakili keadaan lokasi dimana sedimen tersebut diperoleh. Tujuan dari penelitian ini adalah untuk mengetahui keadaan total bakteri, keadaan C/N ratio dalam sedimen dan hubungan antara keduanya terhadap pencemaran sungai Sekembu Jepara. Metode yang digunakan dalam penelitian ini adalah metode deskriptif, untuk pengambilan sampel dilakukan dengan metode purposive sampling . Pengambilan sampel dan pengukuran parameter kualitas sedimen dilakukan di 2 stasiun yaitu pada bagian hulu sungai dan hilir sungai dimana setiap stasiun ditentukan 3 titik sampling. Sedimen dikoleksi dengan sediment sampler berdiameter 6 cm. Total bakteri dihitung dengan metode TPC menggunakan media Nutrient Agar. Jumlah total bakteri sedimen per 1 gram pada sungai Sekembu Jepara berkisar antara 1,6x10 5 – 2,3x10 5 (cfu/g). Nilai C/N ratio pada sedimen di sungai Sekembu Jepara berkisar antara 3,98 – 36,45. Hasil analisis menunjukkan adanya perbedaan nyata jumlah bakteri antara tepian kiri (selatan) dan tepian kanan (utara) sungai Sekembu Jepara dengan nilai p =0,035. Terdapat perbedaan nyata C/N ratio pada sedimen di daerah terkena limbah langsung dan muara sungai Sekembu Jepara dengan nilai p = 0,0303. Tidak ada korelasi yang kuat antara total bakteri dengan C/N ratio pada sedimen di sungai Sekembu Jepara dengan nilai r 0,192. Sungai Sekembu Jepara diduga sudah tercemar oleh berbagai penumpukan limbah domestik maupun limbah industri. River has a potential role for human. The use of sediment as a referance more presicely, because sediment relatively still and not moving like a river water, so it is more representative and representing the state of location where the sediment obatined. The purpose of this research was to find out the total bacteria, C/N ratio and the relationship between the two againts pollution of Sekembu Jepara river. The method used in this research was descriptive method, using purposive a sampling technique. Sampling and measurement of soil and water quality parameters were done at two stations, at upstream and downstream , each station consist ed of three sampling points. Sediment were collected using a sediment sampler diameter 6 cm. The total bacteria was determinated by TPC method using Nutrient Agar. Total bacteria in 1 gr sediment was between 1,6x10 5 – 2,3x10 5 (cfu/g). C/N ratio in the sediment ranged beetwen 3,98 – 36,45. T here was a significant difference of total bacteria in the sediment between the left side (south) and right side (north) of the river with value of p =0,035. T here was a significant difference of C/N ratio in the sediment on upstream and downstream of the river with value of p = 0,0303. There is no high correlation between total bacteria with C/N ratio in the sediment with value of r = 0,192.
